Abstract

Morphine often causes Opioid-Induced Constipation (OIC) due to the activation of μ-opioid receptor, and the activation & translocation of PPARγ which stimulates the AQP3 expression resulting in increased water transport from the intestinal lumen side to the vascular side. The aim of this study was to confirm that the AQP3 is functionally contributes to the onset of morphine-induced constipation using an AQP3 inhibitor (CuSO4) treatment, and to analyze the effect of the laxative osmotic (MgSO4) on PPARγ expression. Mice were divided into four treatment groups: the negative control group, the morphine group, the morphine + CuSO4 group, and the morphine + MgSO4 group. The degree of constipation were evaluated 1h, 2h, and 3h after morphine administration. PPARγ mRNA expression levels were analyzed using the RT-PCR method. The results showed that morphine caused constipation in mice by decreasing the fecal pellet number, weight, and water content. The AQP3 inhibitor agent, CuSO4 was able to inhibit the function of AQP3 in morphine-induced constipation mice by increasing the fecal water content 2h after morphine administration. Meanwhile, the osmotic laxative MgSO4 was able to improve morphine-induced constipation by increasing the fecal water content and the fecal weight 2h after morphine administration. PCR data of PPARγ expression level showed no significant difference among all groups. In conclusion, CuSO4 and MgSO4 relieve morphine-induced constipation by influencing the water transport, but do not affect the frequency of defecation. CuSO4 and MgSO4 do not affect PPARγ mRNA expression.
