Abstract

This study aims to detect the prevalence and pattern of antibiotic resistance in Escherichia coli from broiler chicken sold in the wet market area of East Surabaya, Indonesia which was isolated from 28 June to 27 August 2021. Sampling used the purposive sampling method. The study was conducted in two stages, namely isolation and identification, and antibiotic resistance testing. The isolation stage is a pre-enrichment stage using Buffered Peptone Water (BPW) media, then selective and differential stages are carried out in Eosin Methylene Blue Agar (EMBA) media. The identification stage is biochemical test using Triple Sugar Iron Agar (TSIA), Indole, and Simmons Citrate Agar (SCA) to confirm Escherichia coli. The antibiotic resistance test phase was carried out using the Kirby-Bauer disc diffusion method to determine the resistance of Escherichia coli isolates to Nalidixic Acid (NA), Chloramphenicol (C), Cefotaxime (CTX), Ciprofloxacin (CIP), Meropenem (MEM), and Ampicillin (AMP). The results showed that the overall test of Escherichia coli resistant to NA, C, CTX, CIP, MEM, and AMP was 25 isolates (80.64%) of 31 Escherichia coli isolates. Overall, of the 25 isolates of Escherichia coli that were resistant to antibiotics, 15 isolates were detected as multidrug-resistant (MDR) (48.38%). The conclusion of this study is that broiler chicken meat has the potential as a reservoir of Escherichia coli bacteria that are resistant to various drugs, which can harm public health
