Abstract

Alzheimer's disease (AD) is progressive dementia that affects cognitive function. Inhibition of acetylcholinesterase (AChE) might be a useful therapeutic target for AD. The ethanolic extract of Senna spectabilis leaves is known to have activity as AChE inhibitor. This study aim to determine the effect of 96% ethanolic extract of S. spectabilis leaves on improving cognitive function in scopolamine-induced mice as a model of AD using the Morris Water Maze (MWM) method. In this study, mice were divided into 6 groups. The normal control and negative control were given CMC Na 0,5%, the positive control was given galantamine 4 mg/kg BW, while the extract groups were given 100, 200, and 500 mg/kg BW of extract administered orally 60 minutes before each trial for 7 days. The animal model of AD was induced with scopolamine administered intraperitoneally 30 minutes before trial on day 7. Measurement of cognitive function using MWM was carried out on day 5 to day 9. The results showed that administration of 96% ethanolic extract of S. spectabilis leaves at a dose of 500 mg/kg BW for 7 days independently increases learning ability and spatial memory in mice evaluated by MWM. For further research with the MWM method, other parameters such as path length can be added to ensure the correlation of results. In addition, it is suggested to improve the dose of scopolamine and galantamine to obtain more representative results, while the administration of drugs and extracts should be extended to be able to provide a protective effect.
