Abstract

This study is to investigate the incorporation of the co-processed cushioning agent in Multiple Unit Pellet System (MUPS) to protect Metformin HCl-coated pellets from compaction-induced damage. To confer protection, a cushioning agent should deform more readily than the coated pellet, resulting in a good balance between plastic and brittle deformability. Apart from its profound plastic deformability, an effective cushioning agent should achieve a smooth surface morphology and be smaller than coated pellet to reduce the degree of pellet damage. Cushioning agents containing microcrystalline cellulose (MCC) with Starch 1500® (PGS) or polyvinylpyrrolidone (PVP K 30) in a 1:1 ratio were co-processed by wet beads milling and freeze-drying and compacted with Metformin HCl pellets using 14 mm punch. Both cushioning agents (MCC-PVP and MCC-PGS) exhibited a larger f1 and a smaller f2, indicating a dissimilar dissolution behavior to the uncompacted pellets. The deformation behavior of cushioning agent was analyzed by an Out-die Heckel plot using a 6 mm punch, and the co-processed effect was further characterized by PXRD and optical microscopy. Interestingly, the results obtained from the Heckel plot demonstrated the high plastic deformability of both cushioning agents than untreated MCC and readily deformed at low compression pressure. As plastic deforming excipients (Py<80), no superior cushioning properties were observed from both co-processed cushioning agents in retaining the mechanical integrity of pellet coating during compaction. Hence, further SEM analysis is supplementary to study the surface morphology of cushioning agent and their interparticulate bonding in MUPS tablet.
