Abstract

Hepatitis infection has become a global problem. Therapy with direct-acting antivirals (DAAs) has a high effectiveness but has a risk of resistance and side effect. Medicinal plants have great potential in the development of hepatitis C antiviral drugs. Sida rhombifolia L. is malvaceae's plant family. Previous research stated that the ethanolic extract of the leaves of S. rhombifolia has antiviral activity for hepatitis C with an IC50 value of 2.0 ± 0.6 µg/ml and has hepatoprotective activity. Furthermore, the combination of medicinal plants with current antihepatitis C virus agents show to potentiate their overall effectiveness. Therefore, a combination of hepatitis C antiviral activity was tested for the ethanolic extract of the leaves of S. rhombifolia with antiviral drugs consisting of simeprevir, ribavirin, and daclatasvir. Combination activity of extract S. rhombifolia with simeprevir, ribavirin, and daclatasvir increase antiviral activity. The combination demonstrated synergistic effects which is showing a decreasing IC50 simeprevir by 40 % with a Combination Index value (CI) = 0.446, decreasing IC50 ribavirin by 3 times with a Combination Index value (CI) = 0.646, and decreasing IC50 daclatasvir by 12.1 times with a Combination Index value (CI) = 0.212. In addition, the third combination also increased the value of hepatocyte cell viability when compared to the single sample test. This result suggest that the combination may pottential candidate for anti-HCV complementary or alternavite HCV drugs development.
