Abstract

Background: Bone density is an important factor determining functional bone quality. The mandibular bone density found in individuals with chronic periodontitis was reported significantly lower than that of healthy individuals. However, the mechanism of reduced mandibular bone density in chronic periodontitis has yet to be clearly explained. Objective: The purpose of this study was to explore the mechanism of the disruption of osteoblast proliferation and differentiation on the reduced mandibular density of chronic periodontitis. Methods: 40 male Wistar rats were divided into chronic periodontitis groups which injected with 2×109CFU/mL of P.gingivalis in the sulcular gingiva and control groups which injected with normal saline. After 2, 3, 4 and 6-week injection periods, the subjects were sacrificed and radiographic and micro-Computed Topography (micro-CT) examination of the mandibular bone subsequently performed. The number of osteoblasts, osteoclasts and the expression of TGFβ, HMGB1, Sox2, Runx2, osteocalsin and ALP were evaluated immunohistochemistry. Mandibular bone density was evaluated by histometric analysis. Results: There were a decreased in the mandibular bone density, number of osteoblasts and an increased in osteoclasts number of all periodontitis group. The expression of TGFβ, HMGB1, Sox2, Runx2, osteocalsin and ALP in mandibular bone of periodontitis group were higher than control group in the time-dependent observation. Conclussions: Chronic periodontitis induced P.gingivalis can disrupte the proliferation and differentiation of osteoblasts by increased TGFβ and Sox2 signaling resulting in the reduced of mandibular bone density. This could be use as basic information to be considered in the effort of preventive and comprehensively regenerative based therapeutic of reduced mandibular bone density in chronic periodontitis.
