Abstract

Spilanthes acmella, known as Legetan, is one of Indonesia’s medicinal plant that potential to be an anti-osteoporotic agent because of its sterol and phytotestosterone content. Previous in vitro study showed that butanol and water fraction from this plant increased alkaline phosphatase which is known as a marker of bone formation. The objective of the current study is to analyze anti-osteoporotic effect of 70% ethanol extract, n-hexane, ethyl acetate, butanol, and water fractions of Spilanthes acmella in osteoporosis induced glucocorticoid male mice by increasing their bone density of femoral’s trabecular. This study use 6 month old male healthy mice which induced by dexamethasone for 4 weeks (except the healthy control) and randomizely divided into 7 groups; osteoporotic, control positive (inducted with alendronate), ethanol extract, n-hexane, ethyl acetate, buthanol, and water fractions groups. Each of them were given oral suspension of the corresponding alendronate, extract or fractions for 4 weeks. The results were analyze with Post hoc test LSD and showed that n-hexane fraction (p=0,043) and ethyl acetate fraction (p=0,05) have significant differences with osteoporosis group (p<0,05). It also supported by terpenoids screening using thin layer chromatography method with anisaldehyde-sulphuric acid as TLC stains which gave positive reaction in n-hexane and ethyl acetate fraction. In summary, n-hexane and ethyl acetate fractions have anti-osteoporotic activity by increasing bone density of femoral’s trabecular male mice induced with glucocorticoid.
