Abstract

Pregnancy diagnosis is an important part in reproduction management of ruminant. Pregnancy Associated Glycoprotein (PAG) structurally related to aspartic proteinase, expressed in the outer epithelial cell layer (tropectoderm) of ungulate plecenta. Bovine PAG synthesized by mono- and binucleic trophoblast before complete implantation. Of this, Bovine-Pregnancy Associated Glycoprotein (boPAG) could be used as a marker for early pregnancy. The research on Pregnancy Associated Glycoprotein (PAG), a blastocyst protein isolated from cow pregnant serum was conducted to find out it’s characterisation. In this study, concentration of the Pregnancy Associated Glycoprotein (PAG) were investigated in the milk of dairy cow. The aim of the research was to identification the PAG from Fresian Holstein milk. The procedure included collection milk from pregnant cow in day 90. The cow were diagnosed as pregnant by rectal palpation. After SDS PAGE test, it was ten different molecular weights isolating from cow milk, namely 210,51 ;149,97; 138,89; 102,28; 95,27; 72,63; 59,88; 51,41; 40,25 and 37,85 kDa. After chromatography by Nitroselulose membrane, the PAG molecular weight isolating from Fresian Holstein milk is 59,88 kDa.
