Abstract

This research was aimed to investigate the colour differences between Escherichia coli and Pseudomonas aeruginosa colonies in artificial medium that was made from beef extract, mung beans juice, citrate and bromthymol blue. The media were made by mixing 1.6 % agar base, 0.8 % beef extract, 20 % mung beans extract, 0.2 % sodium citrate, 2 % lactose, 8 % bromthymol blue solution and aquadest ad 1000 ml. The isolate of Escherichia coli and Pseudomonas aeruginosa were inoculated onto the media with five repetitions. As control, Escherichia coli and Pseudomonas aeruginosa were inoculated onto the media that were made from beef extract and mung beans juice, without citrate and bromthymol blue. The whole media were incubated at 37 oC for 24 hours. The result showed that the colour of Escherichia coli colonies can be distinguished with the colour of Pseudomonas aeruginosa colonies. On the treatment media, Escherichia coli colonies appeared colorless, wet, shiny and the surrounding medium remained yellow. The Pseudomonas aeruginosa colonies appeared bluish-white, with thick and mucoid form and the surrounding medium changed its color to green. Based on the result, the artificial medium that was made from beef extract, mung beans juice, citrate and bromthymol blue can be used in Microbiology Laboratories as a differential medium to distinguish between the bacteria whether they utilize citrate as energy and carbon source or not.
