Abstract

This research was aimed to investigate whether the medium made from beef extract, mung beans juice, citrate and bromthymol blue were be able an alternative medium for simmons citrate agar. The media were made by mixing 1.6 % agar base, 0.8 % beef extract, 20 % mung beans extract, 0.2 % sodium citrate, 8 % bromthymol blue solution and aquadest ad 1000 ml. The isolate of Pseudomonas aeruginosa were inoculated into the five media. As control, Pseudomonas aeruginosa were inoculated onto the simmons citrate agar media. The whole media were incubated at 37 oC for 24 hours. the result showed that the colour of the surrounding medium changed the color from yellow to light blue while the colour of simmons citrate agar changed from green to old blue. Based on the results, the artificial medium that was made from beef extract, mung beans juice, citrate and bromthymol blue can’t be used as an alternative medium for simmons citrate agar because the colour of medium different with simmons citrate agar. .
