Abstract
The aim of this study was to isolate and characterize the cellulolytic bacteria from rumen fluid beef cattle. The method used in this study starts with a sampling of rumen fluid beef cattle and then dilute ranging from 10-1 to 10-5, then taken by 1 ml on Carboxy Methyl Cellulose (CMC) media using spread plate method for 24hours incubation with 39°C, then purified on Nutrient Agar media. The observe of cellulolytic activity is using cleared zone diametre and the characteristic of cellulolytic bacteria is using microscope. The results of this study were obtained 10 isolates of bacteria with the bacillus and coccus form, and gramnegative and gram-positive , four of which S3 ( 10mm ) , S6 ( 10mm ) , S9 ( 10mm ) and S10 ( 11mm ) in diametre hallo greater than 6 other bacterial isolates and have optimal cellulolytic activity . The conclusion of this study is there are cellulolytic bacteria in the rumen fluid beef cattle has the form bacillus, gramnegative and hallo 10-11mm diametre.
