Abstract

The aim of this research was knowing the correlation between APAF-1 and apoptotic cells by intrinsic route in the bursa of Fabricius of chicken infected by virulence Gumboro virus type. Twenty-four of commercial strain broiler chickens were 22 days old which dividing two groups randomly. They are twelve chickens as a control (P0) and an other as treatment group (P1). Each of chicken in treatment group was infected by 106 EID50 by Medan, Jawa Barat and Bali isolate of virulence Gumboro virus type through intraocular, intracloacal and oral routes and the control group were given by placebo. Then both of treatment groups were waited until 24 hours post infection. The bursa of Fabricius of all chickens were taken and identified by imunohistochemistry method and hystopatology to APAF-1 expression, but to be identified by S7101 Apopteg Plus Peroxidase In Situ Apoptosis Detection Kit (Tunnel Assay) which used to apoptotic cells. This value was analyzed using t-test (with 5% significant) and SPSS 13 for Windows. The final result show that there was increased number of expression of APAF-1 and apoptotic cells. Both of them were significant difference (p < 0.05) between control and treatment groups (P0: P1). The conclusion of this research showed that there was correlation between APAF-1 and apoptotic cells (intrinsic method) in the bursa of Fabricius of chicken infected by virulence Gumboro virus type.
