Abstract

This study was conducted to understand the influence of swim up time on the motility, viability, and plasma membrane integrity of spermatozoa to meet the best quality of spermatozoa used in In Vitro Fertilization. Ram semen was collected using artificial vagina then fresh semen motility, viability, and plasma membrane integrity were examined. Semen collection was conducted twice a week. In this study sample were divided into four treatment group 0 minute, 30 minute, 60 minute, and 90 minute incubation. After meet the requirement for further study the semen then incubated using BO (Brackett – Oliphant) medium, for every treatment group were added 0.3 semen added 0.6 BO medium then incubated for 0 (zero), 30 (thirty), 60 (sixty), 90 (ninety) minute then were checked motility, and viability. All incubation was performed at room temperature. Plasma membrane integrity was checked using HOS (Hypo-Osmotic Swelling) test 0,1 ml semen added with 1 ml HOS incubated for 30 minute at room temperature for each treatment. Motility examination was done under microscope with 400 magnifications. Viability examination using eosin – negrosin staining then also assessed under microscope examination using 400 magnifications. Incubation up to 60 minute gave the best result not only highest percentage of motility, and viability but also the lowest plasma membrane damage.
