Abstract

The aim of this experiment is to evaluate the inhibitory effect of rhizome extract leaves of Curcuma zedoaria administrated at pre initiation period on 7,12- dimethylbenz(a)anthracene (DMBA) induced rat mammary carcinoma as chemoprevention agent from natural substance to reduce proliferation activity and changes in histopatology. The Sprague dawley rats were divided into four treatment groups. Rhizome extract of Curcuma zedoaria was given every day for 7 weeks in 2 doses, there were 300 mg/kg bw and 750 mg/kg bw. DMBA, which was used to induce carcinogenesis, was given twice a week for five weeks, starting from the third week until the seventh week. Palpation of mammary cancer was started at one week after the last DMBA treatment during 12 weeks. Necropsy was performed at 12 weeks after last DMBA treatment has been terminated to be taken up their mammary gland for microscopic examination using AgNOR and H&E method to analyze the cell proliferation activity and the changes of cell histopatology. Result of this study demonstrated that rhizome extract of Curcuma zedoaria could inhibit the development of DMBA-induced rat mammary cancer by reducing the cell proliferation activity and the changes in cell histopathology in both doses as a blocking agent. It can be concluded that rhizome extract of Curcuma zedoaria with dose 300mg/kg bw are capable to delay the progression of mammary cancer, therefore it should be taken into account for chemopreventing agent in mammary cancer.
