Abstract

The aim of this research is to isolate the tyrosine kinase of dairy cow spermatozoa with Western Blot method and perform antibody specificity test of polyclonal tyrosine kinase with Dot Blot method tyrosine kinase protein that had been isolated from plasma membrane spermatozoa of dairy cow be identified by Western Blot method. For Dot Blot method Five healthy local hare (Oryctolagus cuniculus) was immunized with Tyrosine kinase protein (TK) that had been isolated from plasma membrane of dairy cow spermatozoa. On the day first, each hare received TK + CFA (Complete Freund’s Adjuvant) (1 : 1) dosage 150 μl. Second injection, hares received TK + IFA (Incomplete Freund’s Adjuvant) (1 : 1) dosage 150 μl. Booster had been done at second and seven weeks after second injection using TK + IFA (1 : 1) dosage 150 μl. Blood had been taken 3 ml for each hare and sentrifuged (about 20 minutes using 1500 rpm) to separate the serum. Purification of the serum using SAS 50 % method to get the proteins. This research positively showed tirosine kinase expression with Western Blot method either the specificity of antibody polyclonal of tyrosine kinase using Dot Blot on immunized hare.
