Abstract

Penelitian ini bertujuan untuk mengetahui efek timbal dan kadmium terhadap kadar hemoglobin, nilai Mean Corpuscular Haemoglobin (MCH) dan Mean Corpuscular Haemoglobin Concentration (MCHC) tikus putih (Rattus norvegicus L.). Timbal merupakan logam yang dapat berikatan dengan gugus sulfihidril pada enzim S-ALAD, ferokelatase dan hem sintetase (HS) yang menghambat sintesis hemoglobin. Timbal juga dapat menginduksi terbentuknya oksidan yang akhirnya dapat meningkatkan destruksi eritrosit. Sedangkan kadmium merupakan logam yang dapat menurunkan uptake besi yang menyebabkan penurunan kadar hemoglobin sehingga terjadi hipoksia, menurunkan produksi eritropoietin yang berperan dalam regulasi pembentukan eritrosit dan menyebabkan apoptosis sel darah. Penelitian ini menggunakan 20 ekor tikus putih (Rattus norvegicus L.) betina yang berumur ± 8 minggu (2 bulan). Rancangan percobaan yang digunakan adalah Rancangan Acak Lengkap. Tikus putih dikelompokkan dalam 5 kelompok perlakuan, yaitu kelompok A (diberi 1 ml akuades), B (diberi 1 ml larutan timbal nitrat 50 ppm), C (diberi 1 ml larutan timbal nitrat 100 ppm), D (diberi 1 ml larutan kadmium klorida 50 ppm), dan E (diberi 1 ml larutan kadmium klorida 100 ppm). Sampel darah diambil sebanyak 2 ml dengan cam heart puncture setelah 30 hari perlakuan kemudian diukur kadar hemoglobin, dihitung jumlah eritrosit dan nilai PCV-nya dilanjutkan dengan penghitungan nilai MCH dan nilai MCHC. Data dianalisis dengan menggunakan uji distribusi Kolmogorov-Smirnov, uji Anava (a=0,05), jika ada perbedaan nyata maka dilakukan uji lanjutan yaitu uji Beda Nyata Terkecil (BNT). Hasil penelitian menunjukkan bahwa pemberian timbal dan kadmium secara terpisah menyebabkan penurunan kadar hemoglobin pada semua kelompok perlakuan terutama pada kelompok perlakuan Cd 100 ppm (11,15 g/dl = 22,03 %), menaikkan nilai MCH terutama pada kelompok perlakuan Cd 100 ppm (23,97 pg = 33,17 %), serta menurunkan nilai MCHC terutama pada kelompok perlakuan Cd 100 ppm (25,69 g/dl = 15,10 %). Dari hasil penelitian yang dilakukan pada tikus putih yang diberi perlakuan dengan timbal dan kadmium diatas, dapat disimpulkan bahwa tikus putih mengalami anemia karena penurunan jumlah eritrosit, diduga sebagai efek terjadinya defisiensi besi. Translation: These research were designed to observed the effect of lead and cadmium to the blood of rat (Rattus norvegicus L.) including the haemoglobin concentration, Mean Corpuscular Haemoglobin (MCH) and Mean Corpuscular Haemoglobin Concentration (MCHC). Lead was metal that binding sulphyhidril cluster with S-ALAD, ferrochelatase and haem synthetase (HS) enzyme so could disturb haemoglobin synthesis. Lead also increasing oxidants then erythrocyte cell death were occurred. Meanwhile, cadmium was metal that decreased iron uptake and haemoglobin concentration, then caused hypoxia and decreased production of erythropoietin has role in erythropoiesis regulation and caused apoptosis of blood cell. This research uses 20 female rats with approximately 8 weeks ages (2 months). Design of this research was Complete Random Design. Rats were grouped into 5 treatment group, there were 4 rats in each group. Group A (treated with 1 ml of aquades), B (treated with 1 ml of 50 ppm lead nitrate solution), C (treated with 1 ml of 100 ppm lead nitrate solution), D (treated with 1 ml of 50 ppm cadmium chloride solution), and E (treated with 1 ml of 100 ppm cadmium chloride solution). Blood sample which was taken for 2 ml by heart puncture method after 30 days of treatment then measured of the haemoglobin concentration, number of erythrocyte, PCV, MCH and MCHC. Data were analyzed in Kolmogorov-Smirnov (distribution test), then continued by Anova (a=0,05), and followed by Least Significance Difference (LSD) test if the result were significant. The result of research shows that treatment with lead and cadmium decreasing of haemoglobin concentration especially in Cd 100 ppm (11,15 gldl = 22,03 %) treatment group, increasing of MCH especially in Cd 100 ppm (23,97 pg = 33,17 %) treatment group, and increasing of MCHC especially in Cd 100 ppm (25,69 gfdl = 15,10 %) treatment group.
