Abstract

Menopause adalah suatu kondisi berhentinya menstruasi/haid akibat hilang /kekurangan hormon estrogen. Penurunan estrogen dapat menyebabkan naiknya kadar total lipid, kolesterol LDL serta penurunan kadar HDL. Penelitian ini bertujuan untuk mengetahui peranan ekstrak etanol daun Handeuleum terhadap kadar total lipid, kolesterol LDL dan HDL serum darah mencit yang diovariektomi bilateral. Hewan coba yang digunakan adalah mencit betina dari strain BALB/C dengan umur 2 bulan, berat badan berkisar antara 27-32 gram. Penelitian ini menggunakan rancangan acak lengkap, merupakan penelitian eksperimental murni dengan menggunakan 5 ekor mencit yang tidak diovariektomi bilateral dan 15 ekor mencit yang diovariektomi bilateral. Mencit yang tidak diovariektomi bilateral merupakan kelompok normal (N) dan diberi 0,05 ml aquades. Sedangkan mencit yang diovariektomi bilateral dibagi menjadi 3 kelompok perlakuan, yaitu : kelompok K1 diberi 0,05 ml aquades, kelompok K2 diberi 0,05 ml minyak ikan, kelompok P diberi ekstrak etanol daun Handeuleum dengan konsentrasi 0,5 mg yang dilarutkan pada minyak ikan sebanyak 0,05 ml. Semua perlakuan diberikan secara peroral selama 30 hari. Pengambilan data dilakukan hari berikutnya setelah perlakuan selesai. Parameter penelitian ini adalah perubahan kadar total lipid, kolesterol LDL dan HDL. Data dianalisis dengan ANOVA dan dilanjutkan dengan Uji Beda Nyata Terkecil pada a = 0,05. Hasil penelitian ini menunjukkan bahwa diketahui bahwa akstrak etanol daun Handeuleum dapat menurunkan kadar total lipid serum darah dari 564 mg/dl (K1) menjadi 483 mg/dl (P); menurunkan kadar kolesterol LDL serum darah dari 35,4 mg/dl (K1) menjadi 24,4 mg/di (P); dan tidak berpengaruh terhadap HDL dari 52,4 mg/dl (Kl) menjadi 49,8 mg/dl (P). Kesimpulan dari penelitian ini bahwa pemberian akstrak etanol daun Handeuleum mampu menurunkan kadar total lipid dan kolesterol LDL, serta tidak berpengaruh terhadap kadar HDL. Translation: Menopause is the condition decreasing level of estrogen because the stop of menstruation. The decreasing level of estrogen can increase total lipid also LDL cholesterol and decrease HDL concentration blood serum This research had been conducted to find out the effect of ethanol extract of Handeuleum's leaf to the total lipid, LDL cholesterol and HDL concentration blood serum of ovariectomized mice. Mice were used for this research were female mice from BALB/C strain, age 2 month, and 27-32 gram in weight. This research, which is made by using a randomized-design, was an experimental research that used 5 unovariectomized mice (normal group) and 15 ovariectomized mice which are divided into 3 group as K1, K2, and P. Normal and K1 group were given 0,05 ml aquades, K2 group were given 0,05 ml fish oil. And P group were given 0,5 mg ethanol extract of Handeuleum's leaf in 0,05 ml fish oil. All conductions were orally provided for 30 days. The data was taken after conduction over. The parameter of this research were total lipid, LDL cholesterol and HDL concentration blood serum. The data were analysed by One Way analysis of Variance (ANOVA) and followed by Least Significant Difference (LSD) test. The result designates that the providing ethanol extract of Handeuleum's leaf can decrease the total lipid blood serum from 564 mg/dl (K l) to 483 mg/dl (P); can also decrease LDL cholesterol concentration blood serum from 35,4 mg/dl (K1) to 24,4 mgldl (P); do not influence HDL concentration from 52,4 mg/dl (K I) to 49,8 mg/dl (P). The result of this research showed that ethanol extract of Handeuleum's leaf can decrease the total lipid and LDL cholesterol, and do not influence the HDL concentration of ovariectomized mice.
