Abstract
Penelitian ini bertujuan untuk mengetahui ada tidaknya perbedaan viabilitas, struktur dan perkembangan mikrospora Phalaenopsis amabilis (L.) Bl. pada medium kultur dengan penambahan sumber karbon sukrosa maupun maltosa. Penelitian ini merupakan penelitian eksperimental laboratorik menggunakan Rancangan Acak Lengkap dengan P1 = Medium New Phalaenopsis (NP) dengan sukrosa dan P2 = Medium New Phalaenopsis (NP) dengan maltosa, keduanya dalam inkubasi suhu ruang (280C). Eksplan mikrospora diisolasi dari kuncup bunga dengan panjang 0,8-1,6 cm yang telah mendapat perlakuan suhu rendah (4oC) selama 7 hari dan kemudian dikultur pada medium starvasi dengan inkubasi suhu tinggi (35oC) selama 6 hari. Selanjutnya mikrospora disubkultur pada medium NP dengan penambahan sukrosa dan NP dengan maltosa. Pengambilan data dilakukan sejak awal penanaman dengan perlakuan stres suhu dan medium starvasi sampai pada subkultur ke medium NP (New Phalaenopsis) dengan inkubasi suhu ruang. Hasil penelitian menunjukkan viabilitas mikrospora di medium New Phalaenopsis sukrosa pada awal subkultur sebesar 74,67% menurun pada hari ke-6 hingga 66,58%, sedangkan pada maltosa viabilitas awal sebesar 68,67% menurun pada hari ke-6 menjadi 61,67%. Analisis statistik persentase viabilitas mikrospora menggunakan uji Kolmogorov-Smirnov dan diperoleh p > 0,05 (0,1). Hal ini berarti bahwa tidak ada perbedaan viabilitas mikrospora Phalaenopsis amabilis (L.) Bl. pada medium kultur dengan penambahan sumber karbon berupa sukrosa maupun maltosa. Dari data foto pengamatan diketahui bahwa mikrospora Phalaenopsis amabilis (L.) Bl. dengan penambahan sumber karbon sukrosa dan maltosa pada medium kultur mengalami pembentukan tabung polen dan perkecambahan. Namun, tidak ada perbedaan signifikan antara perkembangan mikrospora di kedua medium, sehingga sukrosa maupun maltosa sama baiknya untuk digunakan sebagai sumber karbon pada medium kultur mikrospora Phalaenopsis amabilis (L.) Bl. Translation: This research was aimed to know there any difference of sucrose and maltose as carbon source addition into New Phalaenoposis medium toward the viability, structure and development of Phalaenopsis amabilis (L.) Bl microspore. This research was an experimental laboratorical research that used Completedly Randomized Design. The treatment were P1 = NP medium with sucrose and P2 = NP with maltose, incubation at room temperature (280C). Microspore explant were isolated from flower bud with length 0.8-1.6 cm put in cold treatment (4oC) for 7 days. The microspores were cultured on starvation medium and incubated at 35oC for 6 days. Afterwards, the microspore were moved into New Phalaenopsis (NP) with sucrose and maltose as carbon source. The data were taken since early planting with stressing temperature and starvation medium treatment until 2 weeks after transferring the microspores into NP (New Phalaenopsis) medium with room temperature incubation. The result showed that early percentage viability at sucrose New Phalaenopsis medium is 74,67% get down to 66,58% on days 6, whereas at the maltose New Phalaenopsis, percentage viability beginning at 68,67% and get down to 61,67% on days 6. Percentage of microspore viability were analyzed with Kolmogorov-Smirnov test and the results was p > 0,05 (0,1), which meant that there was no difference of sucrose and maltose as carbon source addition in New Phalaenopsis medium toward the Phalaenopsis amabilis (L.) Bl microspores viability. On other hand, the data from photograph observation showed that there was pollen formed and germination. However, there was no significant difference between the two medium, either on sucrose or maltose as well as for used in medium of Phalaenopsis amabilis (L.) microspores culture.
