Abstract
Tuberculosis (TB) is a global health issue, and moreover there are increasing cases of multidrug-resistant tuberculosis (MDR-TB) in the world. So the researchers have been trying to find new tuberculosis therapy that is more effective. In this study, the therapy that has been chosen is (-)-epigallocatechingallate (EGCG), from tea leaves (Camellia sinensis), used in combination with each of the first line antituberculosis drugs. The aim of this study is to find whether EGCG can increase the activity of the first line antituberculosis drugs against M. tuberculosis. If EGCG can increase their activity, it can be new alternative tuberculosis treatment that is more effective. The minimum inhibitory concentration (MIC) of each antituberculosis drug was determined by broth dilution method using Middlebrook 7H9 medium, the MIC result was 5 ppm, 0.5 ppm, 50 ppm and 5 ppm for rifampicin, isoniazid, pyrazinamide and ethambutol, respectively. Then the MIC of EGCG combination with each antituberculosis drug was also determined by the same method. The MIC after combination was 0.5 ppm, 0.25 ppm, 20 ppm and 2 ppm for rifampicin, isoniazid, pyrazinamide and ethambutol, respectively. The result shows that the MIC of each first line antituberculosis drug was decreased due to the combination of EGCG. It shows that EGCG gives synergism effect when combined with those drugs, so it can be concluded that EGCG can be used as a complement of antituberculosis drugs for more effective tuberculosis therapy.
