Abstract

Restoration of NiCr can undergo corrosion process in saliva due to corrosion mechanism that is not released from electrochemistry involving transports of electrons. Corrosion product is soluble oxidized substances in salivary electrolytes. The aim of this study was to reveal the mechanism of Ni toxicity as a corrosion product of restoration NiCr recasting on gingival fibroblast through expression analysis of P 53, Bcl-2 and apoptosis. The design of this study was an experimental time series. The samples were immersed in saliva and observed for 1, 3, and 7 days. The highest Ni concentration was exposed to gingival fibroblast culture. The analysis of Ni solubility in artificial saliva was conducted using test of Atomic Absorption Spectrophotometer (AAS), whereas expression of P 53, Bcl-2 and apoptosis was carried out using immunocytochemistry staining. The data collected were subsequently analyzed using two-way analysis of variance, followed by one-way analysis of variance. Comparing between experimental groups after one-way analysis of variance was conducted using Fisher’s LSD (Least Significant Difference). Whereas, the calculation of protein expression of P 53, Bcl-2, and apoptosis of gingival fibroblast was documented using camera of Olympus Microscope BX-50 Japan. Statistical analysis of two-way analysis of variance showed the presence of interaction between recasting frequency and immersion time on Ni solubility in artificial saliva valued p < 0,0001; the interaction between increasing Ni concentration and exposure duration on the expression of gingival fibroblasts P 53 valued p < 0,0001; interaction between the increasing Ni concentration and exposure duration on the expression of Bcl-2 gingival fibroblasts valued p=0,021< =0,05; as well as the interaction between increasing Ni concentration and exposure duration on the apoptosis of gingival fibroblasts valued p < 0,0001. It can be concluded that the higher frequency of recasting, the greater the solubility of Ni, and the higher concentration of Ni exposed to gingival fibroblasts, and the longer incubation time, will increase the expression of P 53 and apoptosis, but decreased Bcl-2 expression. The new finding in this study is the revelation of toxicity mechanism of Ni as NiCr restoration corrosion recasting products through the expression of P 53, Bcl-2 and apoptosis in gingival fibroblasts
