Abstract

Metformin HCl microspheres for oral delivery system is an alternative to deliver metformin HCl which improved efficacy and produced prolonged release. Some factors that affected physical characterization microspheres (produced by ionotropic gelation method with aerozolisation technique using alginate as polymer and CaCl2 as crosslinker) are polymer concentration and crosslinker concentration. The aim of this research was to investigate effect of alginate concentration (1,25% and 1,75%) and crosslinker concentration (3% and 5%) on characteristics of metformin HCl-Ca alginate microspheres. Metformin HCl-Ca alginate microspheres were successfully prepared by ionotropic gelation using aerosolization technique. Microspheres were resuspended in 5% maltodextrin solution as lyoprotectant and were freeze-dried for 29 hours. The microspheres were evaluated for their physico-chemical characteristics such as FT-IR, particle size, morphology, drug loading (DL), entrapment efficiency (EE), and yield. FT-IR showed that there were interactions between drug, polymer, and crosslinked and importantly the microspheres were formed. All mircospheres were spherical, smooth and have partikel size below 10μm. Particle size increased by increasing polymer and CaCl2 concentration. Drug loading decreased by increasing polymer and CaCl2 concentration. Entrapment efficiency increased by increasing polymer concentration and decreasing CaCl2 concentration. Polymer and CaCl2 concentration has no significant differences in yield.
