Abstract

Gastric ulcer is a lesion occurred in the surface of mucosal gastric due to several condition. Stress is one of the cause of the gastric ulcer occurrence even though the mechanism is not clearly discovered yet. One of the mechanism suspected causing the gastric ulcer are the unbalanced protecting and destructive factors of mucosal gastric. The aim of this study is to understand and to compare several SSRI antidepressants effect on stress induced gastric ulcer. The model used for stress was water immersion restrain stress and the drugs used are fluvoxamine 50 mg/kg and 100 mg/kg, fluoxetine 10 mg/kg and 20 mg/kg, paroxetine 10 mg/kg and 20 mg/kg and sertraline 10 mg/kg and 20 mg/kg. The samples then evaluated macroscopically by measuring the ulcer index and intraluminal bleeding score, continued with microscopic observation of the mucosal gastric tissue using Hematoxylin eosin stain. The results showed that each SSRI antidepressant had different antiulcer effect on stress induced gastric ulcer. The mice given fluvoxamine 50 mg/kg and 100 mg/kg, fluoxetine 10 mg/kg and 20 mg/kg, and paroxetine 20 mg/kg significantly decrease the ulcer index induced by stress (p<0,05). Meanwhile the administration of fluoxetine 20 mg/kg and paroxetine 20 mg/kg decrease the intraluminal bleeding (p<0,05).
