Abstract

The aim of this research was to validate High Performance Liquid Chromatography (HPLC) method for simultaneous determination of EGCG, EGC, ECG and EC in green tea product. Validation parameters such as selectivity, linearity, accuracy, precision, LOD and LOQ was observed. Problem was occurred during the study whereas EGC, ECG, and EC didn’t show any absorbance. So, this study focus on validation of High Performance Liquid Chromatography (HPLC) method for simultaneous determination of EGCG and caffeine in green tea product. Calibration curves showed linearity in the range of 40-200 ppm for EGCG and caffeine with r value ≥ 0.999. The percentage recovery of EGCG and caffeine were 98,49% - 99,47% and 97,04% - 102,63%, respectively. One sample t-test value for the percentage recovery of analytes indicated a high accuracy degrees with α = 0.05. The coefficient variance of intraday and interday precision for EGCG were 2,0% and 0,7% and for caffeine were 1,9% and 1,1%. LOD and LOQ for EGCG were 0,45 ppm and 1,52 ppm, also 1,30 ppm and 4,35 ppm for caffeine. This result showed that method meets the validation method requirement. When this method was applied in sample product, the value of EGCG in product was 5,7% b/b with KV 1,7% and the value of caffeine was 8,5% b/b with KV 1,2%.
