Abstract

Fibrinolytic enzymes are serine proteases used for the treatment of cardiovascular disease. The fibrinolytic mechanism is as a remover of thrombus. Many fibrinolytic enzymes are obtained from microorganisms. Bacillus sphaericus BM 9.1 isolated from Mangrove Soil Ecotourism Wonorejo Beach, Surabaya has fibrinolytic activity. Bacillus needs nutrients to grow. KNO3, NH4Cl, NaNO3 and (NH4) 2SO4 are inorganic salt nitrogen sources which are the growth medium of Bacillus sphaericus BM 9.1. This research to determine the effect of inorganic salt nitrogen source on fibrinolytic activity of Bacillus sphaericus BM 9.1. For growth media of Bacilus sphaericus BM 9.1 used Nutrient Agar + Inorganic salt nitrogen source and Agar + inorganic salt nitrogen source with each medium added bacterial suspension 10μL then incubated 121ºC for 24 hours. The nitrogen source used is KNO3, NaNO3, NH4Cl and (NH4)2SO4. Then make Fibrin plate media then perforated with sterile holes with a diameter of 4mm hole, after that the production media perforated with sterile piercing with the same diameter. Then inoculating the production media to Fibrin plate media then incubated 121ºC for 24 hours. Of the four types of inorganic nitrogen sources obtained the highest fibrinolytic activity of KNO3 5,83 ± 0.07. After optimizing the four concentrations of 1%, 2%, 3% and 4%, the highest fibrinolytic enzyme activity at concentration of 3% was 5.88 ± 0.14. Along with increased concentrations also increased nutrients. According to the results of one-way variance analysis it can be concluded that each concentration leads to significant differences in fibrinolytic activity of Bacillus sphaericus BM 9.1
