Abstract

This study aimed to isolate and characterize Pregnancy Associated Glycoprotein (PAG) from pregnant cow blood serum, and to prove that Pregnancy Associated Glycoprotein (PAG) can induce humoral immune response by forming antibodies against Pregnancy Associated Glycoprotein (PAG). PAG was isolated from pregnant cow with 60 days of pregnancy. SDS-PAGE was performed on PAG isolate to obtain PAG isolate with 46 kDa molecular weight. Specificity test was performed to identify specific proteins of PAG using Western Blot method. Polyclonal antibodies to PAG were made by using Two male New Zealand rabbits immunized with PAG isolates. Five cc blood was taken on each rabbit through auricular vein at week 4-8 after the injection of booster for analysis of PAG antibody titre by using indirect ELISA. The Research data of molecular weight of PAG isolate and PAG antibody titer obtained are presented in descriptive form. The results showed that 10 protein bands were identified on SDS-PAGE examination. The molecular weight of the protein are between 10-200 kDa. The Western Blot method showed a visible PAG protein band with a molecular weight of 46 kDa. PAG proteins can induce humoral immune response with the formation of PAG antibodies with OD between 0.305-0.623.
