Abstract

Melicope latifolia (DC.)T.G.Hartley is Indonesian plant which belongs to Rutaceae family and commonly known as Ki Sampang. The previous study reported that the ethanolic extract of M.latifolia (DC.)T.G.Hartley exhibited antiviral activity against Hepatitis C Virus (HCV) JFH1a with IC50 value of 3.5±1.4 μg/ml. The aims of this study were to identify the active compound from ethanolic extract of M.latifolia (DC.)T.G.Hartley leaves collected from Salak Mountain, West Java, Indonesia and determine their antiviral activities against JFH1a. The isolation of ethanolic extract of M.latifolia (DC.)T.G.Hartley leaves was done by several chromatography methods. In vitro anti-HCV assay was performed using hepatocyte cell line (Huh7it) and HCV genotype 2a (JFH1a). Alkaloid quinoline isoprenylated compound, namely N-metilflindersine was isolated from ethanolic extract of M.latifolia (DC.)T.G.Hartley leaves. The structure was determined based on basic spectroscopy analysis. This compound was tested for its anti-HCV against JFH1a. The result showed that Nmethylflindersine was classified as an active compound with IC50 value of 3.24 μg/ml, CC50 value of 100.37 μg/ml and SI value of 30.97. N-methylflindersine might be a potential candidat for anti-HCV drug.
