Abstract

Background: Neuropathic pain due to chronic constriction injury lead to pain behavioral changes and alteration of some substances in dorsal root ganglion and spinal cord. The result of thermal withdrawal latency and immunohistochemical analysis of substances in dorsal root ganglion and spinal cord shows that wet cupping therapy (WCT) has beneficial effects to reduce neuropathic pain in chronic constriction injury (CCI). Obejctive: To evaluate the analgesic effects of WCT against neuropathic pain due to chronic constriction injury in rat dorsal root ganglion and spinal cord. Methods: Four months male Rattus norvegicus rats were divided into five groups, i.e: K0 (negative-controlled), P1 (negative-controlled plus WCT), P2 (Sham-CCI), P3 (CCI) and P4 (CCI plus WCT). WCT two times every week for three weeks with 5 minutes of first cupping using -200 mmHg, ten punctures and 5 minutes of second cupping using -200 mmHg. Results: WCT could reduce neuropathic pain significantly (p<0,05) by increasing the thermal withdrawal latency, reducing P2X3 receptor expression, but significantly increasing opioid-μ receptor, GABA-A receptor, GLT-1 expressions and pNR1 expression (p<0,05). Conclusion: Chronic constriction injury could lead to neuropathic pain. The mechanism of wet cupping therapy to reduce neuropathic pain is through P2X3 receptor, opioid-μ receptor, GABA-A receptor, GLT-1 and pNR1.
