Abstract
This study has a clinical background on sprue infection in HIV-Aids patients, which are thought to be caused by the presence of C.albicans biofilm which colonizes and attacks oral mucous tissue. The discovery of antimicrobial photodynamics that combines light with photosensitizer and oxygen molecules of tissues, has been developed for the purpose of eradicating pathogenic microbes which non-specific and without resistance. The principle of eradicating with antimicrobial photodynamics are through toxicity and reactivity of ROS compounds formed during irradiation. This study aims to find new photosensitizer materials with quantum yield high characteristics. Diffusion of oxygenation in anaerobic biofilms before irradiation aims to increase the effectiveness of chlorophyll photosensitizers in the formation of ROS. The photosensitizer material developed is chlorophyll from papaya leaf extract because it has a high chlorophyll content and contains antifungal substances. The analysis carried out was to determine the chlorophyll characteristics including the maximum absorbs spectrum, quantum yield value and their toxicity to C. albicans biofilm. The data measured were OD490 values, malondialdehyde levels (MDA), profiles of the biofilm cell damage and dead cell accumulation. The maximum absorption of chlorophyll obtained were Soret=414 nm and Q=668 nm with the suitability of the type of laser for irradiation at wavelengths of 445 and 650 nm. The quantum yield of each spectrum is Soret = 0.58 and Q = 0.92. The photoinactivation with a 650 nm laser more effective than a 445 nm laser. The percentage of inactivation at an energy dose of 40 J/cm2 in the biofilm group without oxygenation was 26% for the 445 nm laser and 32% for the 650 nm laser, while the oxygenation biofilm group was 37% for 445 nm laser and 42% for 650 nm laser. Damage profiles on biofilm cells is a shrinking the cell surface, releasing EPS matrix from the cell surface and damage to hyphae cell tissue.
