Abstract

Andrographis paniculata Ness has been known as a medicine for many kind of diseases, one of them is for cancer. To determine the carcinogenesis inhibition effect of A. paniculata extract, it has examined the effect on 7,12- dimethylbenz [a]anthrasene (DMBA)-induced rat lung hyperplasia model in female Sprague-Dawley rats. DMBA 20 mg/kg bw was given twice a week for five weeks by oral. The extract was given in three doses equivalent with andrographolide 10, 30 and 100 mg/kg bw.Carcinogenesis inhibition of A. paniculata extract was evaluated by lung histology that showed decreasing interalveolar septum cells proliferation compared with DMBA group. The role of andrographolide decrease cells proliferation and increase cells apoptosis on lung cancer was evaluated by imunohistochemical methode. Imunohistochemical analysis results that andrographolide can decrease expression of telomerase, an enzyme that contributes to immortality of cells by allowing continued cell proliferation, and increase expression of caspase 3 that considered to be the main executioner caspase in apoptotic process.
