Abstract

Loratadin is a second generation of antihistamin that classified as Biopharmaceutical Classification System (BCS) II, which mean loratadin has low solubility and high permeability. Cocrystal is a method that could improve solubility of loratadin which succesfully able to produced using succinic acid as it’s coformer at a molar ratio of 1:1. In previous studies, cocrystal have been shown to be susceptible to dissociation at high humidity and temperature because of bond release of the two component. This study is observed stability of cocrystal at 30oC / 75% RH, 40oC / 75% RH, 30oC / 90% RH, and 40oC /90% RH. Physical characteristics was investigated using Differential Thermal Analysis (DTA) and Powder X-Ray Diffraction (PXRD). Results from DTA and PXRD showed that dissociation of cocrystal susceptible at 40oC and humidity 90%. Thermogram DTA showed addition endothermic peak of the cocrystal component. PXRD spectrum showed different intensity than cocrystal.
