Abstract
The purpose of this research was to improve a gastroretentive drug delivery system of Rantidine HCl. Ranitidine HCl is a histamine H2 receptor antagonist. Ranitidine is absorbed only in the initial part of small intestine and has 50% absolute bioavailability. Moreover, colonic metabolism of ranitidine is partly responsible for the poor bioavailability of ranitidine from the colon. Hence there is need to develop a dosage form that release the drug in stomach so that it can be absorbed from upper part of gastrointestinal track leading to improved bioavalability. Floating Drug delivery system used to target drug release in the stomach or to the upper part of the intestine. Four different formulas of ranitidine HCl were prepared by wet granulation method using 16% of of hydroxypropyl methylcellulose (HPMC) K100M and HPMC K100LV. The ratio of hydroxypropyl methylcellulose K100M and hydroxypropyl methylcellulose K100LV in FI, FII, FIII, and FIV is 1:0, 12:1, 8:1, 4:1 respectively. The prepared tablets were evaluated on their physical and floating characteristics. The results showed that the physical characteristic tablet was decrease as the increasing concentration of HPMC K100LV in formula’s ratio. Floating lag time, it is a floating lag time tablet increase as the increasing concentration of HPMC K100LV in formula’s ratio. Entirely the formula floats for more than eight hours.
