Abstract

Chloroquine and primaquine are the first-line of anti-malarials drugs that will be prospected combine in tablet. The aim of this study was to develope and validate of method for simultaneous determination of chloroquine and primaquine in tablet formulation by TLC. Validation of method is needed to obtain valid analytical method base on selectivity, linearity, accuracy, and precision parameters. The plate of TLC used Sillica GF254 which had been prior impregnated with KOH 0,1 M. The selected mobile phase was metanol : ammonia (100:1.5). The value of Rf was 0.56 for chloroquine and 0.44 for primaquine with the value of Rs was 4. The selected wavelength was 256 nm. The linearity was good with correlation coefficientsof chloroquine and primaquine were 0.9995 and 0.9992. The Vxo of chloroquine and primaquine were 2.85% and 3.41%. The accuracy was shown by percent recovery. Percent recovery for choloroquine was 93.38  4.85% and 90.07  14.29% for primaquine. The coefficients of variation for choloroquine was 5.19% and 15.87% for primaquine. The result of choloroquine and primaquine determination in sample were 77.52  2.3% and 96,94  3.57%.
