Abstract

Microparticles can be defined as drug delivery systems that have particles size in a range between 1-1000 μm. Ketoprofen, which is included in NSAIDs group has such a short half-lifetime and its frequency of use is often. It is expected to extend the service life of the drug, so the frequency of drug administration can be decreased by formulating ketoprofen as microparticles,. The aim of this research is to investigate the effect of drugpolymer ratio on physical characteristic and release profile of ketoprofenchitosan microparticles. Microparticles was prepared by ionic gelation methods with chitosan 19 cPs as polymer, cross-linked with tripolyphosphate (TPP) and dried by spray drying methods. F1, F2, and F3 each has drug-polymer ratio as 3:15, 5:15, and 6:15. The result showed that microparticles have spherical shape and as the amount of ketoprofen increased, drug content and encapsulation efficiency increased. Release profile of microparticle was tested in stimulated intestinal fluid (pH 6,8) using F2 and F3 as samples. Along with the increase in the number of ketoprofen, there will be a reduction in barriers to the release of ketoprofen microparticles. Release rate of F2 is slower than F3, which could be seen by the slope. F2 and F3 each has slope as 11,7043 ± 0,3305 %/minute 1/2 and 12,3330 ± 0,2747 %/minute 1/2.
