Abstract

Curcumin is a substance obtained from Curcuma longa Linn which has therapeutical effects. Curcumin is insoluble in water at acidic or neutral pH and has poor bioavailability. This problem could be resolved by improving drug dissolution. Solid dispersion is one of the successful method to improve drug dissolution and to obtain better bioavailability. Solid dispersion of curcumin- PEG 800- poloxamer 188 in the ratio 1:2:0,5 and 1:2:1 were prepared with melting method. Differential Thermal Analysis (DTA), powder X-ray diffraction (XRD), fourier transform infrared spectroscopy (FTIR), Scanning Microscope Electron (SEM), and solubility of solid dispersion, physical mixture, and pure curcumin were evaluated. DTA studies revealed that there was a decrease of curcumin endothermic peak. Characterization with XRD showed the significant decrease in cristalinity of pure drug present in solid dispersions or physical mixture. FTIR analysis indicates no interaction between curcumin and PEG 8000, poloxamer 188. SEM photomicrograf of solid dispersions showed that hot melt method were effective in producing highly homogeneous partially solid dispersions systems. Solubility of curcumin solid dispersion showed the better solubility than those observed for physical mixture and pure curcumin.
