Abstract

The aim of this study was designed to determine the characteristics and penetration of diclofenac sodium with w/o microemulsion system from HPMC 4000 gel. The microemulsion contained was surfactant Span80- Tween80 and cosurfactant isopropanol, with ratio 4:1. There was two formula in this study, a gel with w/o microemulsion system as the first formula and gel with w/o emulsion system as the second formula. Characteristics evaluation included organoleptic, pH, ability to spread. The result was showed that microemulsion system had effect on pH, consistency, and spreadibility, but it hadn’t effect on color and odor. The physical stability testing showed that formula I had better stability than formula II. The penetration of diclofenac sodium through Wistar rat skin membrane was determined by dissolution test, which was carried out with apparatus 5-paddle overdisk in phosphate buffer saline 7.40±0.05, temperature 37.0±0.5oC, 100 rpm. The result showed that drug penetration rate for formula I were (0.7144 ± 0.0344) μg/cm2/min and the membrane permeability for formula I were (7.2602x10-5 ± 3.4930x10-6) cm/min. The penetration test tesult from formula I was not compare with formula II because the poor stability of formula II.
