Abstract

Shoot culture of Costus megalobrachtea K.Schum was treated in copper containing media with various concentrations (0; 2,5; 5; 10; 20 and 40 ppm Cu2+). Each group consist of 10 culture bottles and half of them to be cultivated meanwhile the others would be continued to the next passage. Growth index, pH value and % brix of the media would be measured on each passage to see the stability of the shoot culture. The ability of Costus megalobrachtea K.Schum remediate the copper in the media measured by analyzing the Cu2+ residue and its accumulation in the shoot culture biomass with Atomic Absorption Spectrophotometer instrument. The stability of shoot culture of Costus megalobrachtea K.Schum could be seen from the growth index and Cu2+ residue in media for each passage profiles. The accumulation of Cu2+ in Costus megalobrachtea K.Schum biomass was 60,83 ± 5,84 μg/g DW; 125,89 ± 18,33 μg/g DW; 318,11 ± 49,72 μg/g DW; 531,31 ± 27,37 μg/g DW; 1279,99 ± 21,39 μg/g DW for shoot culture group 2,5; 5; 10; 20 and 40 ppm Cu2+ respectively. The highest accumulation was 1279,99 ± 21,39 μg/g or 0.13% w/w DW. This accumulation was higher than 0.1% w/w DW and proved that the shoot culture of Costus megalobrachtea K.Schum has ability to be hyper accumulator plant.
