Abstract

Nanostructured Lipid Carriers (NLC) has been introduced as the second generation of Solid Lipid Nanoparticles (SLN). The main difference between SLN and NLC is performed by nanostructuring the lipid matrix, in order to decrease the particle size and increase the entrapment efficiency. p-Methoxycinnamic acid (PMCA) has irritation effect when administered orally. The aim of this study was to identify the characteristic of PMCA loaded NLC with different lipid ratio of cetyl alcohol (CA) and oleic acid (OA), such as 10,0:0,0; 9,5:0,5; 9,0:1,0; and 8,5:1,5. NLC was prepared using high shear homogenization method at 24000 rpm for 8 minutes. The product were characterized for morphology, particle sized and entrapment efficiency (%EE). As OA ratio increase up to 1,5, the particle size showed the smallest size (276,8 nm) in spherical shape. The % EE of NLC was increase (up to 77,78%) with increasing OA ratio. These result were explained by thermal analysis (DTA) and x-ray pattern (PXRD) study. The increasing of OA ratio to NLC formation resulted crystal lattice disturbance and less ordered matrix of NLC. Hence, increased the % EE of drug.
