Abstract

his study investigated effect of two parameters (polymer concentration and crosslinking time) to optimise the production of ovalbumin-alginate microspheres. This research parameters were sodium alginate concentration of 1,5% w/v and 2,5% w/v and crosslinking time of 30 minutes and 120 minutes. Ovalbumin-alginate microspheres were characterised in terms of physical characteristics (particle size; shape and surface morphology), protein loading, entrapment efficiency, and yield. From 2x2 factorial design of analysis on protein loading, drug entrapment efficiency, and yield at 95% confidence level, showed that sodium alginate concentration had significant effect at drug entrapment efficiency and yield but crosslinking time did not have significant effect at protein loading, drug entrapment efficiency, and yield. Particle size was also influenced by sodium alginate concentration and crosslinking time. Small particle size of ovalbumin-alginate microsphere was produced using this ionotropic gelation-aerosolization method. SEM examination showed spherical shape microspheres with rough surface. Higher concentration of sodium alginate produced higher protein loading, entrapment efficiency, and yield. Whereas, higher crosslinking time tend to lower the protein loading and entrapment efficiency of ovalbumin-alginate microspheres.
