Abstract

Colchicine is an alkaloid from Gloriosa superba L. which has relieve acute gout attacks activity with therapeutic doses of 0.5 to 1 mg daily. Colchicine has a narrow therapeutic index and has a side effect that is quite high, especially at high doses. Therefore, this research is needed to determine the colchicine content in G. superba L. syrup to prevent the incidence of side effects in the use of herbal preparations. Determination of colchicine in G. superba Linn’s syrup using TLC-densitometry method must be validated by the tested parameters selectivity, linearity, precision, accuracy, and range. From selectivity test the mobile phase obtained was chloroform-diethylamine (7:3) with a resolution of 1.77 (> 1.5), linearity test produced regression equation y = 3272.58 + 8748.54x (r = 0.9989) (r table (5%) = 0.7545); Vxo = 2.16%, coefficient of variation of precision 1.44%, and the percent recovery of 86.24% ± 8.83%. While the determination of colchicine in freeze dried leaf infusion G. superba gave 0.2812% ± 5.77% and the determination of colchicine in G. superba syrup of 0.5219 mg/25,0 ml. This method has been validated and can be used in determination of colchicine in G. superba Linn syrup. From this study, 5,68 g or ± 25 fresh leaves of Gloriosa superba L. from Kebun Raya Purwodadi equivalent to 0.5 mg of colchicine can be used in the leaf infusion for public consumption to overcome gout arthritis.
