Abstract

Paracetamol is analgesic drug that is often used. Paracetamol can cause hepatotoxic if it consumed for long time or consumed in high dose. Hepatotoxic is caused by the formation of paracetamol’s metabolite (NAPQI) which bind to liver cell at the orto position of hydroxyl group. The purpose of this study is to synthesize paracetamol derivative (4’-acetamidophenyl-4-methoxybenzoate) and evaluate its analgesic activity. Molecular docking study was also used to predict the analgesic activity by Molegro Virtual Docker and the result showed that the docking score of interaction against cyclooxygenase receptor 2 (3LN1) was lower than paracetamol, therefore that 4’-acetamidophenyl-4-methoxybenzoate was predicted to have greater analgesic activity than paracetamol as a lead. Synthesis 4’-acetamidophenyl-4-methoxybenzoate was carried out by reacting paracetamol with 4-methoxybenzoyl chloride. The TLC and melting point test were indicated the purity of the compound. Structure identification was done based on UV-Vis, FT-IR, and 1H-NMR spectra. The synthesized compound was tested for analgesic activity by hot plate method in mice (Mus musculus) and the result showed that 4’-acetamidophenyl-4-methoxybenzoate has similar analgesic activity with parasetamol.
