Abstract

The purpose of this research to explored antigenic proteins of Leucocytozoon caulleryi as a diagnostic kit candidate of Leucocytozoonosis to broiler chicken. Exploration of antigenic proteins were performed by many steps: identification of gametocyte L.caull eryi from blood examination, isolation of L.caull eryi schizont from chicken organs, fractionation of whole proteins of L.caull eryi schizont by SDS-PAGE assay, and characterization antigenic proteins of L.caulleryi schizont by Western Blot assay. The results showed that the characterization of L.caulleryi schizont protein have been recognized by SDS - PAGE, have molecular weight of 114.7 kDa , 109.6 kDa , 80.5 kDa , 65.0 kDa , 55.6 kDa and 36.1 kDa respectively. Some protein bands that appear thicker seen in protein with a molecular weight of 109.6 kDa , 65.0 kDa and 55.6 kDa. Characterization by Western blot obtained two protein fractions showed thick bands with molecular weight are 109.6 kDa and 65.0 kDa , whereas the other 4 protein fractions do not appear or are not reacted with antibodies ( IgG) from infected chicken serum. The conclusion of the research showed that L.caulleryi schizont has six protein fractions to molecular weight. They were 114.7 kDa , 109.6 kDa , 80.5 kDa , 65.0 kDa , 55.6 kDa and 36.1kDa respectively . Among those protein fractions, 109.6 kDa and 65.0 kDa were antigenic proteins that could be developed as diagnostic kit. Keywords: Leucocytozoon caulleryi, antigenic proteins, broiler chicken
