Abstract
Research of acoustic vibration detection of speckle patterns on a sample of dental plaque biofilm which is an in-vitro study of dental plaque biofilm has been done. The method used for this study was acousto-optic imaging, which are a combination of acoustic system between a ultrasonic and optical system in a single device, in order to obtain speckle imaging. The light modulation caused by the influence of acoustic waves on dental plaque biofilm samples developed on the acrylic medium will cause the speckle modulation carrying physicaJ information from the sample. SignaJs or physicaJ infonnation from samples were obtained by iradiating the laser beam on the sample causing a back-scattering and it was then detected using a CCD camera detector. The results obtained from the detector was recorded on personal computers (PC) in the form of a video image of 320 x 240 pixels, and is displayed in the form of speckle pattern. Speckle pattern was analyzed using software imageJ characterization. The result obtained was the histogram characteristics of gray-level intensity distribution and there was a speckle contrast change on every accoustic vibtration detection by the influence of acoustic vibrations and observation of changes in the formation of dental plaque biofilm samples. For the measurement of the thickness of the sample, as comparison of speckle contrast observations, CLSM equipped with an AOTF was used. The results obtained were as follows: the 2-hour samples with a t 6 J.1ffi thickness has high speckle contrast of0.592707 a.u; the 4-hour samples with a of7 J.1ffi thickness has the speckle contrast of 0.571257 a.u; and the 6-hour samples with a 9 J.Un thickness of has low speckle contrast of 0.550700 a.u. The longer the time required to form dental plaque biofilm the lower the speckle contrast and the thickness of the plaque biofilm samples increases. Whereas, the faster the formation of dental plaque biofil~ the higher the speckle contrast and the thickness of the plaque biofilm samples decreases. There was a change on dental plaque samples obtained from the oral cavity, starting from initial installation stages of plaque biofilm to the formation of mature plaque biofilm
