Abstract

Lead impairs reproductive function because it can induce reactive oxygen species (ROS). ?-tocopherol has an antagonistic effect, which can reduce oxidative stress. Zinc produces an enzyme system that helps neutralise free radicals. This study aims to prove the activity of ?-Tocopherol, and Zinc Sulfate the number of spermatogenic cells (number of spermatogonia, spermatocytes, spermatids) and the number of Leydig cells in white rats (Rattus norvegicus) exposed to lead acetate. The samples used were 25 male Wistar rats with a weight of 150-200 grams divided into five groups. The control group was given aquadest, and corn oil, the P0 group was given lead acetate at a dose of 50mg/KgBW and corn oil. Group P1 was given lead acetate at a dose of 50mg/KgBW and ?-Tocopherol at 100mg/KgBW. Group P2 was given lead acetate at a dose of 50mg/KgBW and zinc sulfate at a dose of 0.54mg/kgBW. Group P3 was given lead acetate at a dose of 50mg/KgBW and a combination of zinc sulfate at a dose of 0.54mg/kgBW and ?-Tocopherol at a dose of 100mg/KgBW orally for 18 days. After the treatment, the rats were sacrificed, and the testes were taken for further histopathological preparations. Observations were made on the seminiferous tubules, including counting the number of spermatogonia, spermatocytes, spermatids, and Leydig cells. Data were analysed using ANOVA continued by Duncan's test with 95% confidence level using SPSS 20 software. The results showed that the combination of ?-Tocopherol and Zinc Sulfate could maintain the number of spermatogenic cells and Leydig cells after exposure to lead acetate.
