Abstract

This research aimed to examine the contamination of Salmonella sp. in chicken meat sold at traditional markets and supermarkets in Karangpilang, Surabaya. Thirty chicken meat samples purchased from each trader in three traditional market and two supermarket was done using random sampling method. Chicken meat then stored in 50 ml Buffered Peptone Water (BPW) and incubated in 37º C for 24 hours. Then, soaked sample stored in tube consist of Selenite Cystine Broth (SCB) and incubated in 37ºC for 24 hours. After being incubated, the soaked samples inoculated with ose on Xylose Lysie Deoxycholate (XLD) agar and incubated in 37ºC for 24 hours. After incubated, cultured samples that suspected as Salmonella sp. taken for biochemical test for futher identification with Triple Sugar Iron Agar (TSIA) media. This treatment and test apply to 30 sample from 5 different markets. The result of conventional way to identify Salmonella sp. show 28 (93,33%) samples on XLD agar and 46 (42,59%) sample on TSIA positive for Salmonella sp. The result of PCR assay shows that 7 (23,33%) samples identified as Salmonella sp.
