Abstract

Amylase is an enzyme that breaks down the starch into dextrin and smaller polymers composed of glucose molecules. This study aims to determine the characteristics of temperature, pH and types of metal ions in increasing the amylase enzyme activity of Bacillus sp. EKA 19. In this study, the bacteria Bacillus sp. EKA 19 endosymbionts of cockroach decomposers as a source of enzymes. This research is included in observational and experimental research with descriptive data analysis. Enzyme activity tests were carried out using the dinitrosalicylic acid (DNS) method and protein levels were measured using the Lowry method. Enzyme production was carried out by incubation for 72 hours. The enzymes produced were collected by centrifugation at a speed of 3500 rpm for 15 minutes. The amylase enzyme from the production has an activity value of 2,498 U/mL and a protein content of 2903,206 mg/mL. The results showed that the enzymes from the production had optimum characters at a temperature of 40 °C and pH 7. The addition of metal ions Ca2+ to the test of the amylase enzyme activity of Bacillus sp. EKA 19 functions as an activator while Fe2+, Cu2+, Mn2+, Mg2+ function as inhibitors.
