Abstract
The aim of this research was to investigate the effect centrifugation, freezing step, and manitol as cryoprotectant on chitosan nanoparticles prepared by ionic gelation then freeze dried. Chitosan nanoparticles that form by ionic gelation method was separated by centrifugation process from its solvent. In this research, the effect of speed and time of centrifugation, freezing temperature and freezing time, and cryoprotectant amount were studied. The result showed that nanoparticles chitosan with mannitol 10% w/v indicated had spherical shape after being centrifuged at 4000 rpm for 5 minutes and freezed at -30oC with 3,5 mbar vacum press for 1 hour. Otherwise, SEM image of dried nanoparticles showed that agregation formed after drying for 24 hours.
