Abstract

This study aims to determine the toxicity effect of chitosan - gelatin - chondroitin sulfate scaffold on BHK 21 and LC50. Treatment with chitosan - chondroitin sulfate and gelatin with A concentration of 35: 65: 0; B 35: 60: 5; C 35: 55: 10; D 35: 50: 15; E 35: 45: 20 which will be made as a scaffold using BHK (Baby Hamster kidney) 21 cells as a sample in each well and incubated for 24 hours. Toxicity results were obtained using the MTT test method and incubated for 4 hours. Optical density absorbance showing cell viability was calculated by ELISA reader with a wavelength of 620nm. Cytotoxicity parameters used LC50 and were analyzed by probit analysis, kruskal wallis and followed by Duncan’s Multiple Range Test (DMRT). scaffold in powder form against BHK (Baby Hamster Kidnay) 21 cells by using MTT assay at a concentration of 100% and 50% in each sample produced that E 50% treatment with a ratio of chitosan (35): chondroitin sulfate (45): Gelatin (20) showed more live cells whereas treatment A 100% with a ratio of chitosan (35): chondroitin sulfate (65): gelatin (0) showed less living cells. However, all treatments produced good living cells because all values showed above LC50.
