Abstract

Citrus nobilis Lour is one of herbal medicine which has anti fertility effect. Citrus nobilis Lour contains vitamin A, vitamin B, vitamin C, hesperidin, limonene, citral, and methylantranilate. Hesperidin is one of the substances in Citrus nobilis Lour which has the function to inhibit the activity of Hyalorunidase enzyme. To be able to use Citrus nobilis Lour as herbal medicine for contraception, toxicity and mutagenic examinations are required to prove that this substance is safe to be used. The configuration of total DNA in spermatozoa is one of the steps of mutagenic examination. Early indication to identify malfunction or mutation in spermatozoa can be seen in the alteration of spermatozoa DNA band profile using agarose electrophoresis method. The aim of this research is to identify spermatozoa DNA band profile of Mus musculus which has been treated by water fraction of Citrus nobilis Lour. In this study, the water fraction of Citrus nobilis Lour was given with doses: 40 mglkg BW; 60 mglkg BW and 80mglkg BW per oral daily to male mice for once time of their spermatogenesis cycle (35 days). The result shows that spermatozoa from control and treatment group have the same total DNA number, 13118 bp. Conclusion: there is no significant change on DNA band profile of spermatozoa between control and treatment group.
