Abstract

Food poisoning in Indonesia based on case reports of extraordinary events in 2017 amounted to 2,401 cases with 576 cases of poisoning due to lead (Pb). The toxic effect of lead on the male reproductive system causes testicular atrophy that affects semen quality, including the number, morphology, motility and viability of sperm. The method used was randomized post test only control group design. This research is using 25 male rats which is randomly divided into 5 groups. The control group (K) was not exposed to antioxidants, P0 was exposed to 50 mg/KgBW lead, P1 was exposed to 50 mg/KgBW lead; vitamin E (?-tocopherol) 100 mg/KgBW. P2 was exposed to 50 mg/KgBW lead and 0.54 mg/KgBW zinc, P3 was exposed to 50 mg/KgBW lead, 100 mg/KgBW vitamin E (?-tocopherol), 0.54 mg/KgBW zinc. After 18 days of treatment, the rats were euthanasia and the epididymal organs were removed for examination of sperm viability and abnormality. The results for control (K), P0, P1, P2, P3 showed a mean of 30.60 ± 4.39; 24.20 ± 4.08; 37.20 ± 4.76; 34.80 ± 3.03; 42.40 ± 2.07 for sperm viability and 9.20 ± 3.70; 11.20 ± 2.77; 8.60 ± 2.07; 6.80 ± 1.92; 4.80 ± 2.77. The data were analyzed using ANOVA test and continued by Duncan's test which showed a significant difference (P < 0.05). In conclusion, the combination of vitamin E (?-tocopherol) 100 mg/KgBW and zinc 0.54 mg/KgBW mg giving repair effect on increasing viability and reducing abnormality of sperm exposed to lead (Pb) acetate.
